Target Information

MAP4K1 (also known as HPK1, Hematopoietic progenitor kinase 1) is a hematopoietic cell-restricted member of the Ste20 serine/threonine kinase superfamily.
MAP4K1 acts as a tissue-specific upstream activator of the MEKK/JNK/SAPK signaling pathway, diminishing T cell receptor (TCR) signaling activity and T cell proliferation by phosphorylating the adaptor protein SLP-76.
JNKs/SAPKs play a crucial role in stress responses in mammalian cells. HPK1 is expressed predominantly in hematopoietic cells, including early progenitors, and specifically activates JNK1 and AP-1-mediated transcriptional activity.
HPK1 binds and phosphorylates MEKK1 directly, and JNK1 activation by HPK1 is inhibited by dominant-negative MEKK1 or MKK4/SEK mutants. Unlike PAK65, HPK1 does not bind Rac1 or Cdc42.
