Product Specific Information
This antibody detects endogenous levels of STAT4 only when phosphorylated at Ser721.
Target Information
STAT4 was originally identified using degenerate primers complementary to sequences encoding conserved regions of other STAT proteins. The STAT4 protein shares high similarity with STAT1 (52%) and STAT3 (47%). Functionally, STAT4 can be tyrosine phosphorylated by Jak1 or Jak2, forming homo- and heterodimers with other STAT family members. Tyrosine-phosphorylated STAT4 binds to the IFN-gamma activated site (GAS), and serine phosphorylation is required for maximal transcriptional activity.
STAT4 expression is mainly restricted to the thymus, spleen, and testis. It is activated by interleukin-12 (IL-12), which is essential for T-cell–independent induction of IFN-gamma and for Th1 differentiation during immune response. STAT4-deficient mice show impaired IL-12–mediated Th1 development and increased Th2 differentiation. Additionally, IFN-α/β–mediated activation of STAT4 is required for IFN-γ production during viral infection.
