This antibody detects endogenous levels of p53 only when phosphorylated at Thr55.

Target Information

The tumor suppressor protein p53 is a sequence-specific transcription factor activated by cellular stress. It mediates cell cycle arrest or apoptosis in response to DNA damage or nucleotide starvation. p53 activation leads to transcription of genes such as p21waf1, contributing to G1 or G2/M arrest. Structurally, p53 includes:

• N-terminal transactivation domain
• Central DNA-binding domain
• Oligomerisation domain
• C-terminal regulatory domain

Phosphorylation at Ser15 is critical for activation and stabilization. p53 blocks proliferation of damaged cells, acts as an anticancer agent, and is associated with aging, oxidative stress, and learning deficits.
Mutations in TP53 are found in various human cancers and hereditary syndromes such as Li-Fraumeni syndrome, leading to loss of tumor suppressor function.
