Product Specific Information
Heat-mediated antigen retrieval is recommended prior to staining, using a 10 mM citrate buffer (pH 6.0) for 10 minutes, followed by cooling at room temperature for 20 minutes.
After antigen retrieval, incubate samples with primary antibody for 30 minutes at room temperature.
A suggested positive control is liver carcinoma tissue.
Target Information
The CASP10 gene encodes a cysteine-aspartic acid protease (caspase) family member involved in apoptosis. Caspases are produced as inactive proenzymes that undergo proteolytic cleavage to form active enzymes.
Caspase-10 cleaves and activates caspases 3 and 7 and is itself processed by caspase 8.
Mutations in this gene are linked to apoptosis defects such as type II autoimmune lymphoproliferative syndrome.
Three alternatively spliced transcript variants encoding different isoforms have been identified.
