Heat-mediated antigen retrieval is recommended prior to staining, using a 10 mM citrate buffer (pH 6.0) for 10 minutes, followed by cooling at room temperature for 20 minutes. After antigen retrieval, incubate samples with the primary antibody for 10 minutes at room temperature.
A suggested positive control is Hodgkin's lymphoma.

Target Information

The protein encoded by the BCL-X gene belongs to the BCL-2 protein family. Members of this family form hetero- or homodimers and act as anti- or pro-apoptotic regulators involved in various cellular activities.
These proteins are located at the outer mitochondrial membrane and regulate the opening of the mitochondrial membrane channel (VDAC). VDAC controls mitochondrial membrane potential and the production of reactive oxygen species, as well as the release of cytochrome C, which are key inducers of apoptosis.
Alternative splicing results in multiple transcript variants encoding two isoforms: the longer isoform acts as an apoptotic inhibitor, and the shorter isoform acts as an apoptotic activator.
