Product Specific Information

The UCHT1 monoclonal antibody reacts with human CD3e, a 20 kDa subunit of the TCR complex. The epsilon chain is essential for proper assembly and surface expression of the TCR complex. CD3 is expressed by thymocytes and all mature T cells. Crosslinking of TCR via immobilized UCHT1 induces activation and proliferation.

Each product contains:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis.
Applications Tested: Pre-titrated and tested on human peripheral blood cells at 5 µL (0.4 µg)/test.
A test is defined as the antibody amount for staining 10⁵–10⁸ cells in 100 µL final volume.

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., PI, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assessment.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining.
  • Add 5 µL per sample to antibody cocktails/master mixes before combining with cells.
  • For single-color controls: use 5 µL per 100 µL cell sample containing 10³–10⁸ cells.

NovaFluor conjugates are based on Phiton technology, utilizing engineered nucleic acid dye structures for optimized fluorescent signatures minimizing spillover and spread.

Target Information

The CD3 subunit complex is crucial for transducing antigen-recognition signals and regulating TCR surface expression. CD3 subunits (gamma, delta, epsilon, zeta) are members of the immunoglobulin superfamily encoded on chromosome 11.
They associate with cytoplasmic signal transduction molecules via tyrosine-based motifs. CD3 plays roles in TCR-induced growth arrest, survival, and proliferation.

Expression Profile:

• Present on 68–82% of normal peripheral blood lymphocytes
• 65–85% of thymocytes
• Expressed in Purkinje cells (cerebellum)
• Not expressed on B or NK cells
• CD3 gene defects are associated with CD3 immunodeficiency.
