Product Specific Information

Description:
The UCHT1 monoclonal antibody reacts with human CD3e, a 20 kDa subunit of the TCR complex. The epsilon chain is required for proper assembly, trafficking, and surface expression of the TCR complex. CD3 is expressed by thymocytes and all mature T cells. Crosslinking via immobilized UCHT1 initiates intracellular signaling leading to activation and proliferation.

Applications Tested:

• Pre-titrated and validated for flow cytometry using human peripheral blood cells.
• Recommended use: 5 µL (0.4 µg)/test in 100 µL final volume.
• Cell number range: 10⁵–10⁸ cells/test.

Compatibility Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., PI, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assessment.

CellBlox Plus Blocking Buffer (Cat. No. C001T06F01):

• Required for optimal staining with NovaFluor conjugates.
• Add 5 µL per sample or per 100 µL of cell suspension (10³–10⁸ cells).
• Recommended to mix with antibody cocktails prior to cell staining.

NovaFluor Technology:

• Based on Phiton nucleic acid dye structures.
• Provides engineered fluorescent signatures with reduced spillover and spread.

Target Information

The CD3 complex transduces antigen-recognition signals into T cells and regulates TCR surface expression. It consists of CD3 gamma, delta, epsilon, and zeta subunits encoded on chromosome 11. These subunits associate with signaling molecules through tyrosine-based motifs. CD3 is expressed on 68–82% of peripheral blood lymphocytes and 65–85% of thymocytes. It is absent on B and NK cells. Defects in CD3 genes are linked to CD3 immunodeficiency.
