Product Specific Information

The UCHT1 monoclonal antibody reacts with human CD3e, a 20 kDa subunit of the TCR complex. The epsilon chain, along with CD3 gamma and delta, is essential for proper assembly and surface expression of the TCR complex. CD3 is expressed by thymocytes and all mature T cells. Crosslinking of TCR via immobilized UCHT1 induces cellular activation and proliferation.

Each product includes:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and validated for flow cytometry on normal human peripheral blood cells.
Use 5 µL (0.4 µg)/test in a final volume of 100 µL. Suitable for 10⁵–10⁸ cells/test.

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., PI, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for best results.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining.
  Add 5 µL per sample before combining with cells.
• For single-color controls, use 5 µL per 100 µL cell sample (10³–10⁸ cells).

NovaFluor conjugates utilize Phiton technology with engineered fluorescent signatures designed to minimize spillover and spread.

Target Information

The CD3 complex transduces antigen-recognition signals and regulates TCR surface expression in T cells. It consists of CD3 gamma, delta, epsilon, and zeta subunits encoded on chromosome 11. These subunits associate with signal transduction molecules via tyrosine-based motifs. CD3 is expressed on 68–82% of normal peripheral blood lymphocytes, 65–85% of thymocytes, and Purkinje cells, but not on B or NK cells. Defects in CD3 genes are linked to immunodeficiency.
