Product Specific Information

The PK136 monoclonal antibody reacts with mouse NK1.1, an antigen expressed by natural killer cells and a subset of T cells in NK1.1 mouse strains such as C57BL and NZB.
Common laboratory strains like BALB/c, SJL, AKR, CBA, C3H, and A do not express NK1.1. For these, use the DX5 (14-5971) antibody.
Simultaneous staining of C57BL/6 spleen cells with PK136 and DX5 shows coexpression in most cells, with smaller DX5⁺PK136⁻ and DX5⁻PK136⁺ populations.

  • Excitation: 405 nm

Super Bright 780 is a tandem dye excited by 405 nm and emits at 780 nm (recommended 780/60 bandpass filter).
Use Super Bright Complete Staining Buffer (SB-4401) when using multiple Super Bright dyes to minimize non-specific interactions.

Compensation Considerations:
When using UltraComp eBeads (01-2222-42) or AbC Total Antibody Compensation beads (A10497), higher compensation values may occur in the violet 450/50 channel. Setting compensation with cells is recommended.

Light Sensitivity:
Protect from light to avoid photo-induced oxidation.

Fixation:
Samples can be stored in IC Fixation Buffer (00-8222) or 1-step Fix/Lyse Solution (00-5333) for up to 3 days at 4°C in the dark with minimal effect on brightness or FRET efficiency. Clone-specific performance should be determined empirically.

NK1.1 (KLRB1, NKR-P1A, CD161)
NK1.1 is a marker for natural killer (NK) cells and some T cell subsets. It belongs to the C-type lectin superfamily and plays a role in NK cell regulation.
The KLRB1 protein is a type II membrane protein with an extracellular C-type lectin domain, transmembrane region, and cytoplasmic tail.
Its receptor, NKR-P1A (CD161), recognizes Lectin-Like Transcript-1 (LLT1) as a functional ligand.
