Product Specific Information
The J105 monoclonal antibody reacts with human PD-1 (programmed death-1), a 55 kDa member of the CD28 immunoglobulin superfamily. PD-1 contains the immunoreceptor tyrosine-based inhibitory motif (ITIM) and plays a key role in peripheral tolerance and autoimmune disease. PD-1 is expressed predominantly on activated T and B lymphocytes. Two ligands, PD-L1 (B7-H1) and PD-L2 (B7-DC), have overlapping functions and are expressed constitutively on some tissues and upregulated on activated antigen-presenting cells.
Costaining experiments suggest that eBioJ105 recognizes a different epitope than MIH4 (Product #11-9969).

Flow cytometry, immunoprecipitation, and immunohistology staining of frozen tissue sections.
Applications Tested:
Tested by flow cytometric analysis of PHA-stimulated normal human peripheral blood cells. Recommended ≤1 µg/test in 100 µL volume. Cell number may range from 10⁵–10⁸ cells/test. Titrate for optimal performance.

Target Information
T-cell activation is regulated by PD-1 (PDCD-1), which mediates inhibitory signals upon binding to its ligands PD-L1 and PD-L2 on antigen-presenting cells. These interactions suppress immune activation in the absence of danger signals. PD-1 knockout mice exhibit hyperactivated immune systems and autoimmune diseases. PD-1 often migrates at higher molecular weight in SDS-PAGE than predicted.
