Target Information
GRP78 is a 78 kDa glucose-regulated protein belonging to the HSP70 family. It resides in the endoplasmic reticulum (ER) and associates with newly synthesized secretory and membrane proteins. GRP78 also interacts with mutant and misfolded proteins, marking them for degradation.
The conserved sequence Lys-Asp-Glu-Leu (KDEL) at the C-terminus ensures ER protein recycling via the KDEL receptor. GRP78 plays a critical role in protein folding, degradation of misfolded proteins, and cell homeostasis, preventing apoptosis.
It serves as a biomarker for hypoglycemia and has neuroprotective functions against glutamate and oxidative stress. Reduced GRP78 levels are observed in Alzheimer’s Disease brains and in cases with PS1 gene mutations. Induction of GRP78 is linked to antitumor drug resistance.
