Target Information

EIF2AK2 (PKR) is one of four kinases that specifically phosphorylate Ser51 of translation initiation factor eIF2-alpha in response to environmental stresses. PKR plays a key role in antiviral defense by activation through double-stranded RNA produced during viral infection. It mediates IFN-induced innate antiviral response, virus-induced apoptosis, and regulation of cell growth and differentiation. PKR induces apoptosis via Fas expression and FADD/Caspase-8 signaling. It can activate both survival and death pathways depending on kinase activity. PKR is involved in multiple signaling pathways, including NF-kappaB, p53, p38, and PDGF. It has implications in tumor suppression and neurodegenerative diseases such as Alzheimer’s, Huntington’s, and ALS. Viruses can inhibit PKR activity, e.g., influenza virus NS1 protein.
