Target Information
The tumor suppressor protein p53 is a sequence-specific transcription factor activated by cellular stress. It mediates cell cycle arrest or apoptosis in response to DNA damage or nucleotide starvation. p53 up-regulation triggers transcription of genes such as p21waf1, leading to G1 or G2/M arrest.
The p53 structure includes:
- N-terminal transactivation domain
- Central DNA-binding domain
- Oligomerisation domain
- C-terminal regulatory domain
Phosphorylation at Ser15 is crucial for activation and stabilization. p53 acts as a tumor suppressor by blocking proliferation of damaged cells. Phosphorylation at Ser392 has been linked to tumor formation. Increased p53 levels are associated with aging, oxidative stress, and learning/memory deficits due to reduced long-term potentiation (LTP).
In normal cells, p53 levels are low, but in many transformed cell lines, expression is high, contributing to malignancy. Mutant forms of p53 fail to bind DNA and lose tumor-suppressive function. Alterations in the TP53 gene occur both somatically in cancers and as germline mutations in cancer-prone families such as those with Li-Fraumeni syndrome.
