Target Information
Cellular oxygen sensor that catalyzes, under normoxic conditions, the post-translational formation of 4-hydroxyproline in hypoxia-inducible factor (HIF) alpha proteins. Hydroxylates specific proline residues in the oxygen-dependent degradation (ODD) domains (NODD and CODD) of HIF1A and HIF2A, with a preference for CODD sites. Hydroxylation enables proteasomal degradation via the von Hippel-Lindau complex. Under hypoxia, hydroxylation is reduced, allowing HIFs to accumulate and activate hypoxia-inducible genes. EGLN3 also regulates apoptosis in cardiomyocytes and neurons, glycolysis via PKM hydroxylation, and DNA damage response by hydroxylating TELO2 to promote ATR/CHK1/p53 pathway activation.
