Description:
Human IL-33 (also called NF-HEV and DVS 27) is a 30 kDa proinflammatory cytokine produced by endothelial and epithelial cells. It is released during necrotic cell death and also found in the nucleus of endothelial cells, functioning as an alarmin or endogenous danger signal. Known alarmins include IL-1α and HMGB1.

IL-33 is the ligand for ST2 (IL-33 receptor), a member of the IL-1 receptor family, expressed on mast cells and Th2 effector T cells, and associated with Th2-mediated inflammation.
Originally thought to be processed by Caspase-1, newer data indicate Caspase-3 or -7 can process IL-33 under physiological conditions.

IL-33 induces IL-5 and IL-13 production, activates basophils, eosinophils, and mast cells, and may also reduce inflammation by blocking IL-1 effects. Human IL-33 shares 55% amino acid sequence identity with mouse IL-33.

Applications Reported:
Human IL-33 Recombinant Protein (Carrier-Free) is biologically active and promotes proliferation of D10S cells in culture.

Applications Tested:
The ED50, measured by proliferation of mouse D10S cells, is ≤ 0.05 ng/mL.

Target Information

IL-33 (Interleukin-33):
A 270 amino acid protein of the IL-1 family with an IL-1-like C-terminal domain. Functions as both a proinflammatory cytokine and an intracellular nuclear factor with transcriptional regulatory properties.

IL-33 binds and signals through IL1RL1/ST2, recruiting MYD88, IRAK1, IRAK4, and TRAF6, activating NF-κB and MAP kinases, and driving TH2 cytokine production (IL-4, IL-5, IL-13).
In vivo, IL-33 induces severe pathological changes in mucosal organs and promotes eosinophilia and splenomegaly.

Highly expressed in high endothelial venules of tonsils, Peyer’s patches, and mesenteric lymph nodes; nearly undetectable in placenta.
Despite its predicted molecular weight, IL-33 often runs higher in SDS-PAGE.
It can co-localize with heterochromatin and act as a transcriptional repressor, supporting its dual role as cytokine and nuclear factor.
