Product Specific Information

This antibody is predicted to react with bovine, canine, chicken, human, mouse, and non-human primate based on 100% sequence homology.
It does not cross-react with other purified substrates of cdk5 (amphiphysin and synapsin).

This antibody is specific for the ~95 kDa Dynamin protein phosphorylated at Ser774 in Western blots of rat brain lysates.
It labels the purified protein phosphorylated in vitro by cdk5 but not by PKC.
Immunolabeling is blocked by the phosphopeptide used as the antigen but not by the corresponding dephosphopeptide.

Store at -20ºC for short term, -80ºC for long term storage.


Target Information

Dynamin is a GTPase required for endocytosis of clathrin-coated vesicles from the membrane.
Its involvement in endocytosis was first discovered in Drosophila melanogaster through the shibire mutation.
Three isoforms exist in mammals: Dynamin1 (neuronal), Dynamin2 (ubiquitous), and Dynamin3 (testes).
Each isoform has splice variants suggesting distinct intracellular targets or vesicle functions.

Common domain features include:

  • N-terminal GTPase domain
  • Central pleckstrin homology domain
  • Proline-rich C-terminal with multiple SH3 domains

These SH3 domains target dynamin to clathrin-coated pits.
Dynamin interacts with AP2 adaptor protein complex and amphiphysin.
