Target Information

ERN1 serine/threonine kinase, also known as endoplasmic reticulum to nucleus signaling 1 (IRE1), is a human homologue of the yeast Ire1 gene product. It possesses intrinsic kinase and endoribonuclease activities and plays a key role in endoplasmic reticulum (ER)-based stress signaling.
IRE1 acts as a sensor of unfolded proteins in the ER and initiates the unfolded protein response (UPR) by splicing XBP1 mRNA, converting it into a potent transcriptional activator.
ERN1 is essential for fetal viability and cell survival under stress conditions that cause unfolded proteins to accumulate in the ER. Depletion or dominant-negative inhibition of IRE1 leads to apoptosis under such stress conditions.
