This antibody has been demonstrated to block activation of monocytes with LPS.


Target Information

TLR4 is a member of the Toll-like receptor (TLR) family, playing a key role in pathogen recognition and activation of innate immunity. TLRs are conserved across species and are structurally and functionally similar.
TLR4 is most abundantly expressed in placenta and in the myelomonocytic subpopulation of leukocytes. It forms part of the LPS receptor complex with CD14 and LY96, mediating signal transduction upon recognition of lipopolysaccharide from gram-negative bacteria.
Mutations in the TLR4 gene influence LPS responsiveness, and multiple transcript variants exist. TLR4 expression is seen in peripheral blood monocytes, some B-cells, and placenta. TLR4-deficient mice show increased susceptibility to Gram-negative sepsis.
