Target Information

TLR4 is a member of the Toll-like receptor (TLR) family, playing a key role in pathogen recognition and activation of innate immunity. TLRs are conserved across species and share structural and functional similarities. TLR4 is highly expressed in placenta and myelomonocytic leukocytes. It mediates cellular responses to lipopolysaccharides (LPS) from Gram-negative bacteria and forms a receptor complex with CD14 and LY96. This receptor facilitates recognition of pathogen-associated molecular patterns (PAMPs) and triggers cytokine production for immune defense. Mutations in TLR4 can affect LPS responsiveness. TLR4 is expressed by monocytes, a subset of B-cells, and placenta. TLR4-deficient mice show increased susceptibility to Gram-negative sepsis.
