Product Specific Information

This antibody reacts with an extracellular epitope of CD22 (BL-CAM), a 130 kDa type I transmembrane glycoprotein (immunoglobulin superfamily) expressed in the cytoplasm of pro-B and pre-B lymphocytes, and on the surface of mature and activated B lymphocytes. It is lost on plasma cells, peripheral blood T lymphocytes, granulocytes, and monocytes.


Target Information

CD22 (BL-CAM) is a type I integral membrane glycoprotein (130–140 kDa) expressed in both cytoplasm and membrane of B-lymphocytes. It appears early in B-cell differentiation, similar to CD19. CD22 membrane expression is limited to mature B cells (CD22⁺) and lost in plasma cells (CD22⁻), useful for phenotyping mature leukemias. Strongly expressed in hairy cell leukemia, and positive in diffuse large B-cell lymphoma and nodular lymphocyte predominance Hodgkin’s lymphoma, but negative in classical Hodgkin’s lymphoma.

CD22 preferentially binds α2,6-linked sialic acid. Ligand-induced tyrosine phosphorylation suggests a regulatory role in B-cell receptor signaling, acting as both a positive regulator via Src kinases and an inhibitory receptor through recruitment of cytoplasmic phosphatases.
