Product Specific Information
This antibody recognizes an extracellular epitope between amino acids 192–697 of CD222 (IGF2 receptor), a ubiquitously expressed 250 kDa multifunctional type I transmembrane protein.
The majority of CD222 is found in the late endosomal/prelysosomal compartment, 5–10% in the plasma membrane, and the truncated (220 kDa) form of CD222 is present in human and bovine serum.
Target Information
Lysosomal enzymes containing one or two mannose 6-phosphate (man6P) moieties are transported within the cell by approximately 300 kDa cation-independent mannose 6-phosphate receptors (CI-MPR).
These receptors function in two interconnected cycles:
- Biosynthetic cycle: Transport of newly synthesized lysosomal enzymes from the trans-Golgi network to late endosomes or early lysosomes.
- Endocytic cycle: Transport of extracellular lysosomal enzymes from the plasma membrane to early endosomes via clathrin-coated vesicles.
The entire pool of MPRs cycles between these compartments every 3 hours. Most MPRs are localized in late endosomes, fewer in the trans-Golgi network, and about 10% at the cell surface.
MPRs also bind type II insulin-like growth factor (IGF-II) in addition to man6P, and a soluble ~240 kDa truncated form circulates in serum, capable of binding both ligands.
