Target Information
KLOTHO is a systemic anti-aging hormone belonging to the glycosidase 1 superfamily. It encodes a type I membrane protein highly expressed in the kidney and brain. In mice, KLOTHO deficiency results in phenotypes resembling human aging, including arteriosclerosis, osteoporosis, skin atrophy, growth retardation, shortened lifespan, and infertility. Overexpression of KLOTHO extends lifespan by inhibiting insulin/IGF1 signaling. It also regulates calcium and phosphorus homeostasis by inhibiting the synthesis of active vitamin D and is identified as a potential tumor suppressor.
