Product Specific Information
The 145-2C11 monoclonal antibody reacts with mouse CD3e, a 20 kDa subunit of the TCR complex. The epsilon chain, along with gamma and delta, is required for proper assembly, trafficking, and surface expression of the TCR complex. CD3 is expressed by thymocytes and all mature T cells. Binding of 145-2C11 to TCR initiates intracellular activation, proliferation, or apoptosis depending on conditions. Commonly used as a phenotypic marker for mouse T cells.
Applications Reported: Flow cytometric analysis
Applications Tested: Tested on mouse thymocytes and splenocytes at ≤1 µg/test (100 µL final volume). Cell numbers may range from 10⁵–10⁸ cells/test. Antibody titration recommended for optimal performance.
Light Sensitivity: Tandem dye sensitive to photo-induced oxidation; protect from light.
Fixation: Samples can be stored in IC Fixation Buffer or 1-step Fix/Lyse Solution for up to 3 days at 4°C in the dark with minimal brightness loss.
Excitation: 488–561 nm
Emission: 775 nm
Laser Compatibility: Blue, Green, Yellow-Green Laser
Filtration: 0.2 µm post-manufacturing filtered.
Target Information
The CD3 complex transduces antigen-recognition signals in T cells and regulates TCR surface expression. It includes CD3 gamma, delta, epsilon, and zeta subunits, encoded by closely linked genes on chromosome 11. CD3 participates in TCR-induced growth arrest, survival, and proliferation. CD3 antigen is expressed on 68–82% of peripheral blood lymphocytes and 65–85% of thymocytes, but not on B or NK cells. Mutations in CD3 genes are associated with immunodeficiency.
