Product Specific Information

Description:
The SK3 monoclonal antibody reacts with human CD4, a 59-kDa cell surface receptor expressed by thymocytes, mature T helper cells, and at low levels on monocytes. CD4 acts as a receptor for HIV, and SK3 blocks HIV binding and mixed lymphocyte reactions. SK3 and RPA-T4 recognize distinct epitopes.

Applications Reported:
Validated for flow cytometric analysis.

Applications Tested:
Pre-titrated for flow cytometry using normal human peripheral blood cells. Recommended use at 5 µL (0.06 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).

Light Sensitivity:
Protect vial and stained samples from light; tandem dye is photo-sensitive.

Fixation:
Samples can be stored in IC Fixation Buffer (Product # 00-822-49) or 1-step Fix/Lyse Solution (Product # 00-5333-54) for up to 3 days at 4°C in dark conditions.

Excitation / Emission / Laser:
488–561 nm excitation; 775 nm emission; compatible with Blue, Green, and Yellow-Green lasers.

Filtration:
0.2 µm post-manufacturing filtered.

Target Information

CD4 antigen facilitates recognition of MHC class II molecules and serves as a co-receptor for HIV. It is primarily expressed on T helper cells, but also on monocytes, macrophages, and dendritic cells. CD4 expression is found in thymus, lymph nodes, tonsils, spleen, and certain non-lymphoid tissues. It plays a role in T-cell activation and may mediate neuronal damage in immune-related CNS diseases.
