Product Specific Information
- For immunohistochemistry (PFA-fixed): incubate antibody for 30 minutes at room temperature.
- Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA, pH 9.0, for 45 minutes at 95°C, followed by cooling at room temperature for 20 minutes.
Target Information
Ki-67 is a nuclear protein expressed during various stages of the cell cycle, particularly in late G1, S, G2, and M phases. It contains a forkhead-associated (FHA) domain that associates with euchromatin at the perichromosomal layer, centromeric heterochromatin, and nucleolus.
Ki-67 shows cell cycle–dependent localization: perinucleolar expression at G1, nuclear matrix expression at G2, and chromosomal association during M phase.
Because it is absent in G0 cells and increases from G1 through mitosis, Ki-67 serves as a reliable proliferation marker. Ki-67 antibodies help determine the proliferative fraction in neoplasms, with prognostic value comparable to thymidine labeling indices. It is also used as a neuronal marker of cell cycling and proliferation.
