Product Specific Information

eBioTY/11.8 recognizes CD73, a 69-kDa GPI-anchored cell-surface protein with ecto-5'-nucleotidase activity.
Expression on myeloid cells (CD11b) is restricted to bone marrow. In humans, CD73 can be induced to secrete a soluble form with IL-2, suggesting a role in activation signaling.
Differences between human and mouse CD73 have been reported — BALB/c mice have more CD4+CD73+ than CD8+CD73+ cells, while the reciprocal is observed in humans.

CD73 is expressed on subsets of lymphocytes and increases during lymphocyte maturation. It is found on memory CD4 T cells and regulatory T cells, involved in immune regulation.

Each product contains:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Suitable for flow cytometric analysis.
Applications Tested: Tested on mouse splenocytes at ≤0.125 µg/test (100 µL final volume). Optimize titration for best performance.

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes such as propidium iodide, 7-AAD, or DAPI.
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining.
  • Add 5 µL per sample before combining with cells.
  • For single-color controls, use 5 µL per 100 µL cell sample (10³–10⁸ cells).

Based on Phiton™ technology using nucleic acid dye structures engineered for optimal fluorescence and minimal spillover/spread.

Laser: 561 nm (Yellow)

Note: NovaFluor Yellow 610 may non-specifically stain B cells in SJL mice.
No non-specific staining observed in BALB/c or C57BL/6 mice.

Target Information

CD73 (Ecto-5'-nucleotidase) catalyzes conversion of purine 5'-mononucleotides to nucleosides (preferentially AMP).
It forms a dimer of two 70 kDa subunits linked via GPI to the plasma membrane.
Used as a marker of lymphocyte differentiation.
Deficiency is associated with immunodeficiency diseases, joint and artery calcification, and intestinal tuberculosis.
Gene localized to 6q14-q21; two transcript variants exist.
