Product Specific Information

Description:
The SHLR17 monoclonal antibody reacts with human interleukin (IL)-17F, a homodimeric 34 kDa cytokine of the IL-17 family (A–F). IL-17F and IL-17A are produced by Th17 cells and have overlapping yet distinct roles in inflammation. IL-17F binding to IL-17RA/IL-17RC induces pro-inflammatory cytokines and chemokines, promoting neutrophil recruitment.

Applications Reported:

• Intracellular staining followed by flow cytometric analysis.

Applications Tested:

• Pre-diluted and tested for intracellular staining of human peripheral blood cells using the Intracellular Fixation & Permeabilization Buffer Set (Product #88-8824-00).
• Recommended use: 5 µL (0.25 µg) per 100 µL test (10⁵–10⁸ cells).

Fluorochrome Information:

• Brilliant Ultra Violet™ 737 (BUV737) emits at 732 nm and is suitable for instruments with a 355 nm UV laser.
• Use Brilliant Stain Buffer™ (Product #00-4409-75) or Super Bright Staining Buffer (Product #SB-4401-42) to minimize polymer dye interactions.
• Protect from light due to photo-oxidation sensitivity.

Fixation and Storage:

• Samples can be stored up to 3 days in IC Fixation Buffer (Product #00-8222-49) or 1-Step Fix/Lyse Solution (Product #00-5333-54) at 4°C in the dark.
• Compatible with short-term methanol-based fixation (≤1 hour).
• Excitation: 355 nm; Emission: 732 nm; Laser: Ultraviolet.

Target Information

IL-17F (Interleukin 17F, CTLA-8):
A cytokine produced by activated T cells, mast cells, and basophils. It induces cytokines, chemokines, and adhesion molecules in epithelial, endothelial, and fibroblast cells. IL-17F functions as a homodimer or IL-17A/F heterodimer, promoting monocyte and neutrophil recruitment and stimulating TNF-α, IL-1β, IL-6, and IL-8 production. It is implicated in chronic inflammatory diseases such as rheumatoid arthritis, psoriasis, and multiple sclerosis. IL-17F also inhibits angiogenesis and induces IL2, TGFB1, and MCP-1 production in endothelial cells.
