Product Specific Information

The 14-4-4S monoclonal antibody reacts with mouse and rat MHC class II, I-E subregion-encoded glycoprotein in strains H-2^k, H-2^d, H-2^p, and H-2^r. Haplotypes H-2^b, H-2^f, and H-2^s do not express I-E antigen.
MHC class II is present on B cells, monocytes, macrophages, and dendritic cells. The 14-4-4S mAb has been reported to block antigen presentation in vitro and cross-reacts with rat.

Applications Tested: Tested on mouse splenocytes; ≤ 0.25 µg/test recommended (100 µL final volume, 10⁵–10⁸ cells/test).

Target Information

MHC class II molecules are transmembrane glycoproteins expressed on professional antigen-presenting cells (macrophages, dendritic cells, B cells). They bind endocytosed exogenous antigens and present them to T cells.
The antigen-binding groove between MHC class II α and β chains is open at both ends and accommodates peptides 15–24 amino acids long.
HLA and MHC antibodies are essential tools in immunopeptidomics for identifying and characterizing neoantigens using HPLC-MS/MS.
