Description:
The M3/84 monoclonal antibody reacts with mouse Mac-3, a 110 kDa protein expressed by mononuclear phagocytes. The Mac-3 antigen is expressed intracellularly by bone marrow monocyte lineage and is upregulated during macrophage differentiation. Peritoneal and tissue macrophages, dendritic cells, and endothelial cells express this antigen on their surface. It is reported that Mac-3 may be identical to CD107b.

Applications Reported:
The M3/84 antibody has been reported for use in flow cytometric analysis.

Applications Tested:
Tested by flow cytometric analysis of mouse bone marrow and thioglycolate-elicited peritoneal exudate cells. Recommended use ≤ 0.5 µg per test (100 µL final volume). Cell number: 10⁵–10⁸ cells/test. Optimize titration for best performance.

Target Information

LAMP2 (Lysosome-associated membrane glycoprotein 2, CD107b) is a member of a family of membrane glycoproteins. LAMP2 provides selectins with carbohydrate ligands, may play a role in tumor cell metastasis, and functions in lysosome protection, maintenance, and adhesion.
Before posttranslational modification, LAMP2 is approximately 46 kDa; mature LAMP2 is 100–130 kDa due to glycosylation.
LAMP2 structure includes:

• Large amino-terminal intra-lysosomal domain
• Hydrophobic transmembrane domain
• Short carboxyl-terminal cytoplasmic tail

LAMP2 is used to monitor chaperone-mediated autophagy and exists in three isoforms: LAMP-2A, -2B, and -2C.
Together with CD107a/LAMP-1, it is a major lysosomal membrane constituent involved in lysosome biogenesis and phagosome fusion.
LAMP2 deficiency leads to autophagosome accumulation, cardiomyopathy, and myopathy (Danon’s disease).
