Product Specific Information

Description:
The O323 monoclonal antibody reacts with human CD27, a lymphocyte-specific member of the TNFR superfamily. CD27 is expressed by a subset of thymocytes and most mature T cells, and is upregulated upon T-cell stimulation. CD27 binds CD70 and mediates T cell–B cell interactions.

Applications Reported:
Suitable for flow cytometric analysis.

Applications Tested:
Pre-titrated and validated for flow cytometry using normal human peripheral blood cells. Recommended usage: 5 µL (0.25 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).

Fluorochrome Information:
Super Bright 600 is a tandem dye excited at 405 nm and emitting at 600 nm. Recommended filter: 610/20 bandpass. Ensure instrument compatibility.

Staining Recommendations:
When using multiple Super Bright dyes, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific interactions.

Light Sensitivity:
Sensitive to photo-induced oxidation. Protect from light.

Fixation:
Samples can be stored in IC Fixation Buffer (Product # 00-8222) or 1-step Fix/Lyse Solution (Product # 00-5333) for up to 3 days at 4°C in the dark.

Target Information

CD27 is a 50 kDa member of the tumor necrosis factor receptor (TNFR) superfamily, including CD40 and CD30. It regulates cell proliferation and apoptosis, and its expression is largely confined to lymphocytes. CD27 forms a disulfide-linked homodimer on thymocytes, peripheral T cells, and some B cells. Upon T-cell activation, CD27 expression increases, and soluble CD27 (sCD27, 28–32 kDa) can be detected in plasma and other fluids. sCD27 serves as a prognostic marker for B cell malignancies. CD27-binding protein (SIVA) is associated with CD27-mediated apoptosis.
Diseases related to CD27 dysfunction include Lymphoproliferative Syndrome 2 and Autosomal Recessive Lymphoproliferative Syndrome.
