Product Specific Information

The BC96 monoclonal antibody reacts with human CD25, the 55 kDa interleukin-2 receptor alpha chain (IL-2Rα).
CD25 is expressed by early progenitors of T and B lineage as well as by activated mature T and B lymphocytes.
By itself, CD25 binds IL-2 only with low affinity; however, association with CD122 (IL-2Rβ) and CD132 (common γ chain) forms the high-affinity IL-2 receptor.
CD25 plays a role in lymphocyte differentiation, activation, and proliferation.

  • A test is defined as the amount of antibody used for staining cells in 100 µL final volume

Fluorochrome Information:
Super Bright 600 is a tandem dye excited with the 405 nm violet laser and emits at 600 nm.
Recommended detection: 610/20 bandpass filter. Ensure instrument compatibility.

Staining Recommendations:
When using multiple Super Bright-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific polymer interactions.

Light Sensitivity:
This dye is sensitive to photo-induced oxidation. Protect samples and vials from light.

Fixation:
Samples can be stored in:

• IC Fixation Buffer (Product # 00-8222)
• 1-step Fix/Lyse Solution (Product # 00-5333)
  for up to 3 days at 2–8°C in the dark with minimal brightness loss.
  Clone-specific performance should be determined empirically.

Target Information

CD25 (IL2 receptor alpha chain/IL2RA) is a cytokine receptor involved in T and B lymphocyte proliferation.
The IL2 receptor complex consists of IL2RA, IL2RB, and IL2RG chains, forming a high-affinity receptor.
Homodimeric IL2RA or IL2RB forms lower-affinity receptors.
IL2 is primarily produced by mature T cells and acts as a growth and differentiation factor, regulating immune responses.
Mutations in IL2RA are associated with interleukin-2 receptor alpha deficiency and immune dysregulation.
