Target Information
KIT (c-KIT)는 MGF(mast cell growth factor, stem cell factor)의 type 3 transmembrane receptor로 작용하는 원암유전자(proto-oncogene)입니다.
KIT는 다양한 조혈 및 비조혈 세포의 성장과 활성화를 조절하며, KIT 유전자 변이는 위장관 기질종양(GIST), 비만세포질환, 급성골수성백혈병(AML), piebaldism 등과 관련되어 있습니다.
특히 D816V 활성 루프 변이에 의한 KIT 수용체의 비정상적 활성화는 비만세포종양 치료 전략의 주요 표적입니다.
c-Kit은 조혈줄기세포, 생식세포, 비만세포, 위장관의 Cajal 세포 등에서 발현되며, SCF(stem cell factor) 결합 시 수용체 이합체화 및 자가 인산화가 일어나 다양한 신호전달 경로(PI3K, PLC-γ, JAK/STAT 등)를 활성화합니다.
c-Kit 수용체 활성은 조혈, 줄기세포 유지, 생식세포 형성에 필수적입니다.
