Product Specific Information

Description:
The OKT3 monoclonal antibody recognizes the epsilon subunit of the human CD3 complex. It exhibits potent immunosuppressive properties and has been effective in treating renal, heart, and liver allograft rejection. CD3 subunits (gamma, delta, epsilon) are essential for TCR complex assembly and expression. CD3 is expressed by thymocytes and all mature T cells. Crosslinking of TCR triggers intracellular activation and proliferation pathways.

Applications Reported:
Validated for flow cytometric analysis.

Applications Tested:
Pre-titrated for flow cytometry using normal human peripheral blood cells. Recommended use: 5 µL (0.125 µg) per test (100 µL final volume). Suitable for 10⁵–10⁸ cells/test.

Fluorophore Information:
APC-eFluor® 780 emits at 780 nm and is excited by the red laser (633 nm). Ensure instrument compatibility.

Light Sensitivity:
This tandem dye is sensitive to photo-induced oxidation. Protect from light.

Fixation:
Samples can be stored up to 3 days at 4°C in the dark using:

• IC Fixation Buffer (Product #00-8222)
• 1-step Fix/Lyse Solution (Product #00-5333)

Excitation / Emission / Laser:
633–647 nm / 780 nm / Red Laser

Filtration:
0.2 µm post-manufacturing filtered.

Target Information

The CD3 complex transduces antigen-recognition signals in T cells and regulates TCR surface expression. CD3 subunits (gamma, delta, epsilon, zeta) are members of the immunoglobulin superfamily encoded on chromosome 11. They associate with cytoplasmic signaling molecules via tyrosine-based motifs. CD3 is present on 68–82% of peripheral blood lymphocytes and 65–85% of thymocytes but absent on B or NK cells. CD3 defects are linked to immunodeficiency.
