Product Specific Information

Description:
The 17A2 monoclonal antibody reacts with the mouse CD3 complex. CD3 subunits gamma, delta, and epsilon are required for proper assembly, trafficking, and surface expression of the TCR complex. CD3 is expressed by thymocytes in a developmentally regulated manner and by all mature T cells. Binding of 17A2 to CD3 initiates intracellular biochemical pathways resulting in cellular activation and proliferation.

Applications Reported:
This antibody has been reported for use in flow cytometric analysis.

Applications Tested:
Tested by flow cytometric analysis of mouse splenocytes. Recommended use ≤ 1 µg/test (100 µL final volume, 10⁵–10⁸ cells/test). Optimize titration for best performance.

Fluorophore Details:
APC-eFluor 780 emits at 780 nm and is excited with the Red laser (633 nm). Ensure instrument compatibility.

Light Sensitivity:
Sensitive to photo-induced oxidation. Protect vial and samples from light.

Fixation:
Samples can be stored in IC Fixation Buffer (Product # 00-8222) or 1-step Fix/Lyse Solution (Product # 00-5333) for up to 3 days at 4°C in the dark with minimal impact on brightness and FRET efficiency.

Excitation / Emission / Laser:
633–647 nm excitation, 780 nm emission, Red Laser.

Filtration:
0.2 µm post-manufacturing filtered.

Target Information

The CD3 subunit complex is crucial for transducing antigen-recognition signals into T cells and regulating TCR surface expression. CD3 gamma, delta, epsilon, and zeta subunits are structurally related members of the immunoglobulin superfamily encoded by genes on human chromosome 11.
CD3 is expressed on 68–82% of normal peripheral blood lymphocytes, 65–85% of thymocytes, and Purkinje cells in the cerebellum, but not on B or NK cells.
Defects in CD3 genes are associated with CD3 immunodeficiency.
