Product Specific Information

The WM-53 monoclonal antibody reacts with human CD33, also known as GP67 and P67, a 67 kDa type I transmembrane glycoprotein in the Siglec (sialic acid-binding Ig superfamily lectin) family. It is specific to the hematopoietic compartment and expressed on monocytes, activated T cells, granulocytes, myeloid progenitors, and mast cells.

Super Bright 436 is excited with a 405 nm violet laser and emits at 436 nm. Use a 450/50 bandpass filter or equivalent.
For multiple Super Bright dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific interactions.

Target Information

CD33 is a transmembrane protein of the Siglec family containing immunoreceptor tyrosine-based inhibitory motifs (ITIMs) that recruit SHP-1 and SHP-2 phosphatases for signal regulation. It binds sialic acid residues in N- and O-glycans and serves as a therapeutic target for acute myeloid leukemia (AML). CD33 is expressed on myelomonocytic lineage cells, granulocyte and macrophage precursors in bone marrow, and peripheral monocytes, but not on pluripotent stem cells. It is a useful marker to distinguish myelogenous leukemia from lymphoid or erythroid leukemias.
