Description:
The P67.6 monoclonal antibody reacts with human CD33 (also known as GP67 and P67), a 67 kDa type I transmembrane glycoprotein belonging to the Siglec family. CD33 is expressed on monocytes, activated T cells, granulocytes, myeloid progenitors, and mast cells, and is highly specific to the hematopoietic compartment.

Applications Reported:
This antibody is suitable for flow cytometric analysis.

Applications Tested:
Pre-diluted and validated for flow cytometry on normal human peripheral blood cells. Recommended usage: 5 µL (0.06 µg) per test in a final volume of 100 µL. Cell count range: 10⁵–10⁸ cells/test.

Fluorochrome Information:
Super Bright 436 can be excited with the violet laser (405 nm) and emits at 436 nm. Recommended filter: 450/50 bandpass or equivalent. Ensure instrument compatibility.

Buffer Recommendation:
When using multiple Super Bright dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401) to reduce non-specific polymer interactions.

Target Information

CD33 is a transmembrane protein of the Siglec family containing immunoreceptor tyrosine-based inhibitory motifs (ITIMs). It recruits SHP-1 and SHP-2 phosphatases, mediating inhibitory signaling and receptor turnover. CD33 is expressed on myelomonocytic lineage cells, including granulocyte and macrophage precursors, but not on pluripotent stem cells. It serves as a marker for distinguishing myelogenous leukemia cells and is a therapeutic target for acute myeloid leukemia. Diseases associated with CD33 dysfunction include gallbladder lymphoma and extracutaneous mastocytoma.
