Target Information
The c-Myc protein is a 62 kDa transcription factor encoded by the c-Myc gene on human chromosome 8q24. It is commonly activated in various tumor cells and plays a critical role in cellular proliferation, differentiation, apoptosis, and cell cycle progression.
Phosphorylation of c-Myc at Thr58/Ser62 by glycogen synthase kinase 3, cyclin-dependent kinase, ERK2, and JNK has been associated with cell proliferation and regulation.
Studies indicate that c-Myc is essential for vasculogenesis and angiogenesis in tumor development. The c-myc oncogene (p62 c-myc) regulates normal cellular proliferation and differentiation, while deregulated expression can induce apoptosis.
Antibodies against c-Myc epitopes recognize overexpressed proteins containing Myc epitope tags fused to either amino- or carboxy-termini of target proteins. c-Myc-, N-Myc-, and L-Myc-encoded proteins function in cell proliferation, differentiation, and neoplastic diseases.
Amplification of the c-Myc gene has been observed in human tumors such as lung, breast, and colon carcinomas, whereas N-Myc amplification is found in neuroblastomas. Translocation of the c-myc locus on chromosome 8 to immunoglobulin loci on chromosomes 14, 2, or 22 is characteristic of Burkitt’s lymphoma and other B-cell lymphoproliferative disorders.
Aberrant c-myc expression occurs in tumors of multiple origins, including colorectal, gastric, gallbladder, hepatic, mammary, ovarian, endometrial, head and neck, pulmonary, prostatic, thyroidal, oral, ocular, nasopharyngeal, endocrine, and hematopoietic neoplasms.
