Description:
The 4D10.1 monoclonal antibody recognizes human Syk, a 72-kDa non-receptor protein tyrosine kinase of the Syk/ZAP-70 family. Syk is highly expressed in B cells, with lower levels in thymocytes, mast cells, and platelets, and can be detected in fibroblasts, epithelial cells, hepatocytes, endothelial, and neuronal cells.
Syk is crucial in B and T cell antigen receptor signaling, activated by phosphorylation upon ITAM recruitment, and phosphorylates downstream proteins such as PLCγ and BLNK/SLP-65. Abnormal Syk expression is associated with tumor cell migration and invasion.

Applications Reported:

• Intracellular staining followed by flow cytometric analysis.

Applications Tested:

• Pre-titrated and validated for intracellular staining and flow cytometry of human peripheral blood cells using Foxp3/Transcription Factor Staining Buffer Set (Product #00-5523-00).
• Recommended use: 5 µL (0.125 µg) per test (100 µL final volume).
• Applicable for 10⁵–10⁸ cells/test.

Target Information

Spleen tyrosine kinase (SYK) and ZAP70 are cytoplasmic tyrosine kinases involved in allergic and autoimmune disease pathways and may play roles in cancer.
Upon activation by BCR or FcεRI, Syk translocates to the plasma membrane and phosphorylates adaptor proteins, initiating downstream signaling such as calcium mobilization, cytoskeleton remodeling, and gene transcription.
Syk binds via its SH2 domains to phosphorylated ITAM motifs, and is further activated by Src-family kinase phosphorylation.
