Product Specific Information

The eBio1-21 antibody reacts with human T cell leukemia/lymphoma 1 (TCL1), a 14 kDa proto-oncogene involved in intracellular regulation of T cell signaling. TCL1 is located at the 14q32.1 chromosome breakpoint region in T-cell prolymphocytic leukemia (T-PLL) and is overexpressed due to chromosomal rearrangements that juxtapose it with T-cell receptor promoter elements.

TCL1 interacts with the pleckstrin homology domain of Akt family proteins, enhancing Akt dimerization and activity, which increases phosphorylation of key signaling substrates such as Ikk complex, mTOR, BAD, p70S6 kinase, FOXO transcription factors, and GSK3β. These substrates regulate cell differentiation, growth, survival, and metabolism.

TCL1 is normally expressed in specific subsets of thymic precursors, plasmacytoid dendritic cells, activated T cells, and B cells up to the germinal center stage. Aberrant expression is associated with T cell malignancies and chronic lymphocytic leukemia (CLL).

Target Information

Overexpression of TCL1 in humans is linked to mature T cell leukemia through chromosomal rearrangements that bring TCL1 near TCR regulatory elements. In normal T cells, TCL1 is expressed in CD4-/CD8- cells but not in later differentiation stages. TCL1 acts as a coactivator of the cell survival kinase AKT.
