Antibody detects endogenous levels of IKB alpha only when phosphorylated at Tyr42.

Target Information

IkB-alpha is a 40 kDa protein that functions to inhibit NF-kappaB activity through protein-protein interaction with NF-kappaB dimers in the cytosol. This interaction masks the nuclear localization sequence of NF-kappaB, preventing its translocation to the nucleus. Various stimuli such as TNF-alpha, IL-1 beta, chemokines, PMA, growth factors, LPS, UV irradiation, viral infection, and other stresses can activate NF-kappaB by inducing degradation of IkB-alpha.
In humans, the gene is located on the q arm of chromosome 14. Activation of NFkB requires phosphorylation of IkB on specific serine residues (Ser32 and Ser36), leading to its degradation. IKK alpha (CHUK) phosphorylates IkB-alpha at these sites, and its activity is stimulated by NFkB-inducing kinase (NIK). The IKK complex consists of IKK alpha, IKK beta, and IKK gamma, each essential for IkB phosphorylation and NFkB activation.
