Product Specific Information
- This antibody does not react with Mouse or Rat.
- For immunohistochemistry (PFA fixed): incubate antibody for 30 min at room temperature.
- Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA, pH 9.0, for 45 min at 95°C, followed by cooling at room temperature for 20 min.
Target Information
The tumor suppressor protein p53 is a sequence-specific transcription factor activated by cellular stress. It mediates cell cycle arrest or apoptosis in response to DNA damage or pyrimidine nucleotide starvation. Upon activation, p53 induces transcription of genes such as p21waf1, regulating G1 or G2/M arrest.
p53 structure includes:
- N-terminal transactivation domain
- Central DNA-binding domain
- Oligomerisation domain
- C-terminal regulatory domain
Phosphorylation at Ser15 is crucial for activation and stabilization, while Ser392 phosphorylation correlates with tumor formation. p53 plays a role in preventing proliferation of damaged cells, acting as an anticancer agent, and is linked to aging and oxidative stress. Elevated p53 levels are associated with deficits in long-term potentiation (LTP) in learning and memory.
Mutations in p53, common in various cancers, impair DNA binding and tumor suppressor function. TP53 gene alterations occur as both somatic and germline mutations, including in Li-Fraumeni syndrome.
