Product Specific Information

The TN3-19.12 monoclonal antibody reacts with mouse, rat, and rabbit tumor necrosis factor alpha (TNF alpha), but not with human TNF alpha. The TN3-19.12 antibody is a neutralizing antibody. Mouse TNF alpha is a 17 kDa factor produced by macrophages, monocytes, neutrophils, CD4+ T cells, and NK cells. A 26 kDa form of TNF alpha is expressed as a membrane-bound molecule. TNF alpha is cytolytic and plays an important role in immune regulation. Dimers and trimers of TNF alpha have been observed.

Applications Reported: Suitable for intracellular staining followed by flow cytometric analysis.
Applications Tested: Tested by intracellular staining and flow cytometric analysis of stimulated mouse splenocytes. Recommended usage ≤ 0.5 µg/test (100 µL final volume). Cell number may range from 10⁵ to 10⁸ cells/test. Antibody should be titrated for optimal performance.

eFluor® 660 is a replacement for Alexa Fluor® 647, emits at 659 nm, and is excited with the red laser (633 nm). Ensure instrument compatibility.

Target Information

TNF alpha is a multifunctional proinflammatory cytokine belonging to the TNF superfamily. Secreted primarily by macrophages, it binds to receptors TNFRSF1A/TNFR1 and TNFRSF1B/TNFBR. TNF alpha regulates immune cells, proliferation, differentiation, apoptosis, lipid metabolism, and coagulation. It exists as multimers but shows a single 17 kDa band under non-reducing SDS-PAGE conditions. Knockout studies in mice suggest neuroprotective roles and tumor necrosis induction. TNF alpha is involved in immune responses to bacterial, viral, parasitic, and fungal infections, as well as tumor necrosis. It is synergistic with interferon-gamma in cytotoxicity and implicated in autoimmune diseases, insulin resistance, and cancer.
