The eBio18F10 antibody reacts with mouse IL-17F, a 37 kDa homodimeric cytokine of the IL-17 family and a Th17 signature marker. IL-17F shares 50% amino acid identity with IL-17A, and both genes are co-regulated during Th17 differentiation. Expression of IL-17F and IL-17A is observed in activated human CD4⁺ T cells and is associated with autoimmune diseases such as rheumatoid arthritis, multiple sclerosis, psoriasis, inflammatory bowel disease, and asthma.
IL-17F induces expression of IL-6, IL-8, GRO-α, ENA-78, TGF-β, MCP-1, G-CSF, GM-CSF, and ICAM-1 in epithelial and endothelial cells. It forms both homodimers and biologically active IL-17A/F heterodimers. IL-17A is most potent in inducing GRO-α secretion, followed by IL-17A/F and IL-17F.
Applications Reported: Intracellular staining followed by flow cytometric analysis.
Applications Tested: Tested on Th17-polarized mouse splenocytes; recommended ≤0.25 µg/test (100 µL final volume, 10⁵–10⁸ cells/test).
Fluorochrome Information: eFluor® 660 replaces Alexa Fluor® 647. Excitation 633–647 nm; Emission 668 nm; Red laser compatible.

Target Information
IL-17F (Interleukin 17F, CTLA-8) is a cytokine produced by activated T cells, mast cells, and basophils. It plays a role in allergic airway inflammation and induces cytokines, chemokines, and adhesion molecules in epithelial, endothelial, and fibroblast cells. IL-17F acts as a homodimer or heterodimer with IL-17A, binding to IL-17R to recruit monocytes and neutrophils. It stimulates TNF-α, IL-1β, IL-6, and IL-8 production, and is implicated in chronic inflammatory diseases such as rheumatoid arthritis, psoriasis, and multiple sclerosis. TGF-β and IL-23 are required for Th17 cell induction and maintenance. IL-17F can inhibit angiogenesis and induce IL-2, TGF-β, and MCP-1 in endothelial cells.
