Target Information
Specialized cells at the midline, which separates the left and right halves of the CNS, play key roles in directing growth cone behavior. In vertebrates, insects, and C. elegans, midline cells produce guidance cues such as nectins and slit, which act as attractants and repellents. Robo proteins act as receptors for Slit and are conserved from fruit flies to mammals. Robo1 and Robo2 are human homologs of the Drosophila Roundabout protein, with Robo1 also homologous to the C. elegans gene sax3 and Robo2 to the zebrafish gene astray.
