Product Specific Information

The DREG-56 monoclonal antibody reacts with human CD62L, a 76 kDa member of the selectin family. CD62L is expressed by neutrophils, monocytes, and subsets of T, B, and NK cells. It binds glycosylated, fucosylated, and sulfated sialylated glycoproteins such as CD34, glycam-1, and MAdCAM-1, mediating lymphocyte rolling on endothelium and homing to high endothelial venules (HEV) in peripheral lymphoid tissues.

Applications Reported:
This antibody is validated for flow cytometric analysis.

Applications Tested:
Pre-titrated and tested by flow cytometry using normal human peripheral blood cells. Use at 5 µL (0.125 µg) per 100 µL cell sample (10⁵–10⁸ cells/test).

Fluorochrome Information:
PerCP-eFluor® 710 emits at 710 nm and is excited with a 488 nm blue laser. Recommended filter: 710/50 bandpass (695/40 acceptable alternative).

Fixation:
Samples can be stored up to 3 days at 4°C in the dark using:

• IC Fixation Buffer (Product #00-8222-49)
• 1-step Fix/Lyse Solution (Product #00-5333-54)

Optical Properties:

Target Information

Human CD62L (L-Selectin) is a 74–95 kDa glycoprotein adhesion receptor. It includes:

• C-type lectin binding domain
• EGF-like domain
• Two short consensus repeat (SCR) sequences
• Transmembrane and cytoplasmic regions

Constitutively expressed on most leukocytes, including lymphocytes (except many memory T cells), monocytes, and polymorphonuclear cells.
