Target Information
CD33 is a transmembrane protein belonging to the sialic acid-binding immunoglobulin-like lectin (Siglec) family. It contains immunoreceptor tyrosine-based inhibitory motifs (ITIMs) that recruit protein tyrosine phosphatases SHP-1 and SHP-2, and is subject to ubiquitin-mediated receptor removal.
CD33 is expressed on cells of the myelomonocytic lineage, binds sialic acid residues in N- and O-glycans, and serves as a therapeutic target for acute myeloid leukemia. It is found on granulocyte and macrophage precursors in bone marrow, but not on pluripotent stem cells. CD33 is also expressed on peripheral monocytes and is a useful marker for distinguishing myelogenous leukemia cells from lymphoid or erythroid leukemias.
Diseases associated with CD33 dysfunction include gallbladder lymphoma and extracutaneous mastocytoma.
