Product Specific Information

This LUVNKLA monoclonal antibody recognizes human and mouse STAT3 phosphorylated on tyrosine 705 (Y705). STAT proteins are transcription factors involved in apoptosis, differentiation, and proliferation, activated by cytokine receptor–associated JAK kinases.
After phosphorylation, STAT3 translocates to the nucleus to regulate gene transcription. STAT3 is activated by cytokines such as interferons, IL-5, IL-6, IL-10, and LIF, and plays a critical role in Th17 differentiation and cell survival. Mutations affecting STAT3 are linked to immune dysfunction and cancer progression.

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-diluted and validated for flow cytometry using stimulated human peripheral blood cells at 5 µL/test (0.06 µg). A test corresponds to 100 µL final volume and 10⁵–10⁸ cells/test.

PerCP-eFluor™ 710 emits at 710 nm (excited by blue laser, 488 nm). Recommended filter: 710/50 bandpass; alternative: 695/40 bandpass. Ensure instrument compatibility.

Target Information

STAT3 is a member of the STAT family of transcription factors activated by receptor-associated kinases in response to cytokines and growth factors. It requires phosphorylation at Tyr705 by JAK1/JAK2 for dimerization and nuclear translocation.
STAT3 interacts with coactivators such as CREB-binding protein and transcription factors c-Jun and CREB. It mediates expression of genes involved in growth and apoptosis. Rac1 regulates STAT3 activity, while PIAS3 inhibits it.
Three alternatively spliced isoforms have been described. STAT3 gene is located on chromosome 17q. Knockout of STAT3 in mice is embryonically lethal.
