Product Specific Information

This CHI2S4N monoclonal antibody recognizes human and mouse STAT6 when phosphorylated on tyrosine 641. STAT6 is activated by IL-4 through phosphorylation by JAK1 and JAK3, promoting T cell proliferation and B cell IgE production.

Specificity was confirmed by ELISA and flow cytometry.
Reported application: intracellular staining followed by flow cytometric analysis.
Tested application: intracellular staining of stimulated human peripheral blood cells at 5 µL (0.03 µg) per test (100 µL final volume).

Recommended protocols:

• Protocol C: Two-step (Fixation/Methanol)
• Protocol A/B: Not suitable for this antibody

Fluorophore Information:
PerCP-eFluor® 710 emits at 710 nm and is excited at 488 nm.
Recommended filter: 710/50 bandpass (alternative: 695/40).
Light-sensitive; protect vial and samples from light.

Fixation:
Samples can be stored in IC Fixation Buffer (Product #00-8222) or 1-step Fix/Lyse Solution (Product #00-5333) for up to 3 days at 4°C in the dark.

Target Information

STAT6 is a transcription factor activated by cytokines such as IL-4 and IL-13. STAT6-deficient cells show impaired Th2 differentiation, reduced IgE class switching, and diminished IL-4-mediated proliferation. STAT6 mRNA is expressed in various tissues including lymphocytes, colon, ovary, spleen, and lung. It exists in two isoforms: STAT6a and STAT6c, and plays a key role in Th2 immune response regulation.
