Description:
SRBCZX monoclonal antibody recognizes STAT5 phosphorylated on tyrosine 694. STAT proteins are activated by receptor-ligand binding that triggers JAK kinase phosphorylation. Phosphorylated STAT5 translocates to the nucleus to regulate gene transcription in a cytokine-dependent manner.
Cytokines such as IL-2, IL-7, and IL-15 activate STAT5 via JAK1/JAK3, while IL-3, IL-5, and GM-CSF act via JAK2. Phosphorylation at Tyr694 is essential for STAT5 dimerization, nuclear translocation, and DNA binding.

Specificity of SRBCZX clone was validated by ELISA and flow cytometry.

Applications Reported:
Intracellular staining followed by flow cytometric analysis.

Applications Tested:
Pre-titrated and validated for intracellular staining of stimulated human PBMCs. Use 5 µL (0.06 µg) per 100 µL test volume. Recommended cell count: 10⁵–10⁸ cells/test.

Staining Protocol:
Use Protocol C (Two-step: Fixation/Methanol). Protocols A and B are not applicable. Detailed procedures available in BestProtocols® – Staining Intracellular Antigens for Flow Cytometry.

Fluorochrome Information:
PerCP-eFluor® 710 emits at 710 nm and is excited by the 488 nm blue laser. Recommended filter: 710/50 bandpass (695/40 acceptable). Protect from light due to photo-oxidation sensitivity.

Fixation:
Samples can be stored up to 3 days at 4°C in the dark using:

• IC Fixation Buffer (#00-8222)
• 1-step Fix/Lyse Solution (#00-5333)

Excitation: 488 nm
Emission: 710 nm
Laser: Blue Laser
Filtration: 0.2 µm post-manufacturing filtered

Target Information

STAT5 alpha is a transcription factor activated by cytokines and growth factors. Upon phosphorylation, it forms dimers that translocate to the nucleus to regulate gene expression.
It mediates responses to IL2, IL3, IL7, GM-CSF, erythropoietin, and growth hormones. Constitutive activation (e.g., via TEL/JAK2 fusion) contributes to tumorigenesis.
STAT5 alpha induces BCL2L1/BCL-X(L) expression, conferring anti-apoptotic properties. STAT5 alpha and beta share 96% sequence identity and are co-expressed in many tissues, with differential expression in muscle and mammary glands.
STAT5 alpha is essential in reproduction, lactation, immune function, and growth regulation.
