Product Specific Information

The SIDI8BEE monoclonal antibody reacts with human and rhesus macaque CD8 beta. The CD8b chain associates with the CD8a chain to form the CD8ab heterodimer expressed on thymocytes and cytotoxic T cells. The CD8 receptor complex binds to MHC class I and is involved in T cell development and activation.

SIDI8BEE detects CD8 beta in both CD8ab heterodimer and CD8bb homodimer, and is cross-reactive with human and rhesus macaque CD8b.

Each product includes:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-diluted and tested on normal human peripheral blood cells (5 µL per test). A test corresponds to 100 µL final volume with 10⁵–10⁸ cells/test.

Note: NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI). Use Invitrogen LIVE/DEAD Fixable Dead Cell stains instead.

CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining with NovaFluor conjugates.

• Add 5 µL per sample to antibody cocktails/master mixes before combining with cells.
• For single-color controls: 5 µL per 100 µL of cell sample (10³–10⁸ cells).

NovaFluor conjugates utilize Phiton™ technology, employing nucleic acid dye structures that enable engineered fluorescence with reduced spillover and spread.

Target Information

The CD8B antigen is a cell surface glycoprotein expressed on cytotoxic T lymphocytes, facilitating cell-cell interactions within the immune system. Acting as a coreceptor with the T-cell receptor, CD8 recognizes antigens presented by APCs in the context of class I MHC molecules.

The functional coreceptor may be a homodimer (αα) or heterodimer (αβ). Both chains share homology with immunoglobulin variable light chains. The CD8B gene encodes beta chain isoforms, with multiple alternatively spliced variants and a related pseudogene on chromosome 2.
